Refractive index matching has been long used in biology (Barer 1955) to measure absorption spectra of small particles that otherwise are distorted by the effect of light scattering. It has also been used to determine the refractive index of small particles, such as bacteria (see Immersion refractometry).

Tuchin (2005) has recently reviewed the potential and limitations of this technique as applied to optical clearing of tissue for in vivo spectroscopy and imaging.
